Isolation, Screening, and Identification of Local Bacterial Isolates Producing Bio-Cellulose

Read More The current study aims to easily, quickly, and cheaply select the local bacterial isolate that produces bacterial cellulose. Ten samples of spoiled fruits (triplicate for each) were taken in Al-Kut City/ Wasit Province/ Iraq. A modified H-S medium was used to isolate the local bacteria from spoiled fruit samples. One-hundred and twelve bacterial isolates were obtained from all samples, where the lemon sample give the highest number of isolates, as it reached 20 isolates, while the banana sample gives the lowest number of isolates reached to 6 isolates. The two steps of screening were achieved to select the best cellulose-producing local isolate, results from the primary screening indicated that only 18 isolates appeared a clear ability to create clear zones as an indicator to produce bacterial cellulose, and the isolates TELE8, TELE11, TEKI4, and TEAP5 gave the highest clear zones of 7.5, 6.1, 6.2, and 6.5 cm, respectively. Also, the second screening step confirmed the results collected from the first step, as the results hydrogel test indicated that only 18 strains showed the ability to produce bacterial cellulose, while the other isolates were unable to do so, as the results showed that the isolate TELE8 was superior in its ability to produce bacterial cellulose than isolates TEK11, TEKI4, and TEAP5. Therefore, the local isolate TELE8 was chosen as the best isolate that produces bacterial cellulose. Thereafter, the phenotype and genotype characteristics were used to identify the TELE8 isolate; its results indicated that the isolate belongs to Komagataeibacter xylinum (previously named first Acetobacter xylinum, and then Gluconacetobacter xylinus). The three methods (static, shaker, and fermenter) were used to produce the bacterial cellulose by using the best local bacterial isolate Acetobacter xylinum TELE8 in HS medium. The results revealed that the highest production of BC (22.8 g/200 ml medium) was obtained when using the fermenter method, while the shaker method did not give the productivity of bacterial cellulose.

Read More Sepsis is an upregulated systemic inflammatory reaction that has been identified as a global health concern. It can lead to multiple organ toxicity, including cardiac, hepatic, and renal dysfunction. Sepsis can result in cardiomyocyte impairment, hypertrophy, and heart failure by increasing oxidative stress and the expression of pro-inflammatory cytokines. Furthermore, elevated cardiac troponin-I (cTn-I) during sepsis impairs heart contractile performance by decreasing myofilament response to calcium. The current study aimed to examine the possible effects of levosimendan against sepsis-induced cardiotoxicity. Forty male mice 8-12 weeks old and weighing 25-30 grams. After two weeks of acclimation, the mice were separated into four groups (n = 10): (1) Healthy group (n = 10). (2) CLP group: CLP operation was performed on mice. (3) DMSO group (4) Levosimendan group: 10 mg/kg intraperitoneally (IP) in 2 divided doses for 5 consecutive days. All groups underwent the CLP procedure on the fourth day, were sacrificed on the fifth day, and then samples were taken. The Levosimendan group exhibited a significant (p < 0.05) decrease in myocardial troponin-I concentration compared to the CLP group. In addition, the inflammatory cytokines (TNF-α, IL-6, and IL-1β) serum levels in the levosimendan group were significantly (p < 0.05) lower than in the CLP group. In addition, the levosimendan group demonstrated a significant (p0.05) increase in myocardial SOD activity and a decrease in MDA level compared to the CLP mice. Histopathologically, the levosimendan group demonstrated a statistically significant (p0.05) reduction in cardiac tissue damage. Levosimendan attenuates sepsis-induced cardiotoxicity via multiple protective effects, including anti-inflammatory and antioxidant effects, according to the findings of the present study. In addition, the direct cardiac protective effect via the decreased cardiac troponin-I level and the attenuation of histopathological alterations during sepsis-induced cardiotoxicity.

Among glycolysis enzymes, lactate dehydrogenase (LDH) is an exciting possible pharmacological target for the cancer treatment. Drawing a comparison between diabetes and cancer, the importance of the excessive lactate generation in the diabetes-cancer interaction should not be overlooked. Therefore, this research was designed to determine the connection between lactic dehydrogenase enzyme (LDH) and the blood glucose in serum samples of women with breast cancer-associated to those with breast cancer and type 2 diabetes mellitus (T2DM).Samples and methods: Seventy-five samples were categorized into three equal groups; group 1: Non-breast cancer or diabetes (control subjects C), group 2: patients suffering from breast cancer (BC)‎, and group 3: patients suffering from breast cancer (BC) and type 2 diabetes mellitus (BC and T2DM), in all samples, the amounts of the total LDH and glucose were measured.Results: The LDH level increased significantly (p≤ 0.001) in (BC and T2DM) group and (BC) group in comparison with the control group (C), respectively, and a significant increase in LDH‎ (p≤0.01) ‎ was obtained in (BC and T2DM) group versus the (BC) group. In addition, compared with the control group, the results indicated that the BC and BC and T2DM groups had the higher levels of glucose and LDH.Conclusion: The LDH is a useful diagnostic marker for the metabolic syndrome and can be used to investigate the metabolic changes linked to breast cancer and diabetic complications. Many years ago, it was known that increased LDH is a poor predictor of cancer outcome. LDH may have a prognostic effect on breast cancer metastases, but this is unclear. Concerning the results of this investigation, we speculate that LDH could act as a predictor of breast cancer. Increasingly, the data suggest that diabetes mellitus is linked to an increased risk of cancer and a significant cause of death in cancer patients.

Read More Background: Fetal weight is an important factor in assessing fetus growth, and arranging for delivery. Different parameters and different models were used to estimate fetal weight, and the 3-dimensional ultrasound plays a crucial role in many aspects, including assessing the fetus's weight.Aim of the study: To precisely assess third-trimester ultrasound using fetal limb cross-section area by 3D ultrasound to predict birth weight of the fetus versus sonographic parameters for predicted fetal weight by two-dimensional ultrasound.Materials and methods: This Prospective-cohort study recruited one hundred pregnant women with a normal pregnancy in their late third trimester. They were taken from the antenatal clinic and had their fetal weights estimated by two-dimensional ultrasound by Modified Hadlock (MH) and three-dimensional ultrasound by measuring the thickness of the soft tissue of thigh, assessment of fetal weight, and follow up to delivery to measure the actual birth weight of the infant and comparing the results. A study was done in the department of Obstetrics and Gynecology at Al-Yarmouk Teaching Hospital.The Ethical Committee code was obtained after getting the scientific approval.Results: One hundred pregnant women were enrolled in this study, their mean age of the women was ‎‎32.6 years old, their mean BMI was 25.6 kg/m2, and most of them had a cephalic ‎presentation (92%); in the current study, Modified Hadlock overestimate fetal weight 159.9 g, while midthigh soft tissue thickness (MTSTT) underestimates the fetal weight - 67.3 gm. Modified Hadlock was non significantly different from zero. In terms of systemic errors, there was a non-significant difference between MH from zero, whereas MTSTT significantly differs from MH., and in terms of absolute error, the 3D model was significantly different from MH. Conclusion: ‎This study found that compared with using the traditional Hadlock's formula, the fetal thigh soft tissue thickness measured by the novel method of three-dimensional ultrasonography is more precisely and specifically related to the antenatal anticipated birth weight.
