Read More The aim of this study is to investigate the antibacterial effect of silver nanoparticles (AgNPs) against multidrug-resistant Salmonella typhi recovered from blood, stool, and fluid specimens. In vitro the antimicrobial activity of silver nanoparticles against S.typhi isolates done by determination of silver nanoparticles activity by agar wells diffusion assay, MIC, sub MIC, and by Microtiter plate (Submic). By agar wells diffusion assay and MIC, the antimicrobial activity of silver nanoparticles showed an inhibition zone, among 50 strains of S.typhi. Five Submic showed that sensitivity was higher among Submic 1, 2, and 3 than those in Submic 4 and 5 that show higher resistance (96%, 94%, and 86% vs. 4%), respectively. By Microtiter plate when we used three diluted concentrations of silver nanoparticles of Submic 1, 2, and 3, significant were found that after 24 hours was significantly higher than that after 48 hours, as Submic 1 (0.19874 ± 0.034156 vs. 0.14864 ±0.025908), respectively with mean differences of 0.050100 (t= 9.794, df: 49, P=0.000) and the means of Submic 2 and 3 was (0.20776 ± 0.031197 vs. 0.15730 ± 0.027060) (t=10.401, df: 49, P= 0.000) and (0.21464 ± 0.030793 vs. 0.16802 ± 0.026111) with mean differences of 0.050460 and 0.046620, respectively and (t= 9.031, df: 49, P= 0.000, respectively. Antibacterial activity of silver nanoparticles has been demonstrated in several investigations, but the reported MIC values range through a wide extent of variation. In our study, silver nanoparticles showed a good antibacterial activity against all the tested pathogens. The results of MIC and Submic tests revealed a higher MIC value for S.typhi compared with the other tested pathogens. It is possible that AgNPs act similarly to the antimicrobial agents used for the treatment of MDR S.typhi infections, which show four different mechanisms of action including: interference with cell wall synthesis, inhibition of protein synthesis, interference with nucleic acid synthesis, and inhibition of the metabolic pathway.

Read More Background: In response to microbial infection and cellular injury, the NLRP3 inflammasome, an essential part of the innate immune system, causes caspase-1 activation and the release of the proinflammatory cytokines IL-1 β and IL-18. However, various inflammatory diseases, including periodontal disease, have been linked to the abnormal activation of the NLRP3 inflammasome.Aim of study: This study was conducted to investigate the crucial role of NLRP3 inflammasome activation and IL-18 in the pathogenesis of periodontitis patients with type 2 diabetes mellitus.Materials and Methods: This case control study included eighty-five participants., their age range (23-55) years, they divided into three groups: two groups had periodontitis one of them with type two diabetic mellitus and the other one systemically healthy, the third group was the control group with clinically healthy periodontium and systemically healthy. Serum samples from both patients and controls were analyzed using an ELISA kit designed to detect NLRP3 inflammasome activity and interleukin-18.Results: Conclusions: Elevated NLRP3 and IL-18 levels in PD +T2DM patients, as well as their positive correlation with clinical parameters, suggest a critical role in the pathogenesis of PD with and without diabetes.. A significant positive correlation also noticed between NLRP3 and each of (GI, PPD, CAL and BOP) in PD+T2DM patients. Regarding the group of PD patients there was no significance correlation between NLRP3 and clinical parameters. Additionally, IL-18 was discovered to have a positive correlation with GI, PPD, CAL, and BOP in PD+T2DM patients, w. Meanwhile, there is a significant correlation between NLRP3 as well as interleukin-18 in patients with and without type 2 diabetic mellitus.Conclusions: Elevated NLRP3 and IL-18 levels in PD +T2DM patients, as well as their positive correlation with clinical parameters, suggest a critical role in the pathogenesis of PD with and without diabetes.

Read More Hypᴏthyrᴏidism is a cᴏmmᴏn metabolic disordеr that influеncеs thе functiᴏn ᴏf many ᴏrgans including the seminal vesicle which plays a key role in male fertility process. This work aimed to evaluate the effects of selenium element on seminal vesicle of hypothyroid male rat. Fᴏrty malе albinᴏ rats wеrе used in this study, which were classifiеd intᴏ 4 grᴏups (10 animals in each group): Group I (control) received nothing. Group II received selenium in a dose of 10 µg/ b.w). Group III (hypothyroid) was given carbimazole in a dose of 6 mg/ kg b.w orally by gastric tube once daily for 3 months to induce hypothyroid state. In group IV (hypothyroid + selenium) the animals were treated by carbimazole as same as group III then selenium was added in last 2 months in a dose of 10 µg/ b.w. At the end of the experiment, blood samples were taken for the assessment of serum TSH, T4 and male hormone (testosterone), then animals were sacrificed, their seminal vesicle was removed and fixed in 10% formalin solution, and paraffin sections were prepared and stained by histological and immunhistochemical staining to be examined under the light microscope. Sеrum cᴏncеntratiᴏn ᴏf TSH significantly incrеasеd whilе thе lеvеl ᴏf T4 and tеstᴏstеrᴏnе cᴏncеntratiᴏns significantly dеcrеasеd in grᴏups III followed by group IV. Hypᴏthyrᴏidism causеd rеductiᴏn ᴏf sеcrеtᴏry prᴏduct, atrᴏphy ᴏf mucᴏsal fᴏlds and vacuᴏlar dеgеnеratiᴏn ᴏf thе sеcrеtary cеlls of sеminal vеsiclе, whilе thе immunhistᴏchеmical study shᴏwеd low rеgulatiᴏn ᴏf androgen receptor in thе cеlls ᴏf this gland. Hᴏwеvеr, thе rats ᴏf grᴏup IV shᴏwеd a significant imprᴏvеmеnt in hᴏrmᴏnal lеvеl and histᴏlᴏgical structurе ᴏf thе sеminal vеsiclе tissuе cᴏmparеd with grᴏup II. So, this study indicated that selenium improved male fertility in hypothyroid state.
